Hi Vets, Is this dark mark on a 16 week puppy’s tongue  cause for concern?

Hello!  Looking from my computer screen alone and contemplating the worrying end of the spectrum, this could be a wound (although it doesn't look like one) or local necrosis (cells dying off), or even some kind of cancer.  It could also be normal discolouration, which I would expect to be completely flat and continuous with the pinker skin of the tongue.  In my opinion, it's a good idea to let your vet check.  If they can't be sure, they may feel that a biopsy is necessary.  All the best and please do let us know how you get on.

On second viewing, having gone back to the picture, I think that the lesion may look raised to me and therefore would reiterate that a vet appointment is definitely a good idea.
